What is VIP?

A synthetic form (aviptadil) has been investigated clinically for pulmonary hypertension, ARDS, sarcoidosis and critical COVID-19, typically by inhalation to exploit its short plasma half-life. In research, VIP is a key tool for studying VPAC1/VPAC2 receptor signaling, circadian rhythm regulation in the suprachiasmatic nucleus, and immune tolerance - it shifts immune responses toward anti-inflammatory Th2/Treg profiles. Its extreme plasma instability (half-life of about a minute) is the central experimental challenge.

Mechanism of Action (Research Context)

Agonist at VPAC1 and VPAC2 Gs-coupled receptors, raising cAMP. Produces vasodilation, bronchodilation, increased secretion, and immunomodulation (suppressed Th1 responses, induced regulatory T cells). Degraded rapidly by neutral endopeptidases.
