SS-31 is a mitochondrial research compound with a molecular weight of 639.8 g/mol and a research half-life of ~2-4 hours. SS-31 (elamipretide) is a synthetic tetrapeptide from the Szeto-Schiller series, engineered to selectively target the inner mitochondrial membrane. Its alternating aromatic-cationic motif lets it cross cell membranes freely and concentrate in mitochondria, where it binds cardiolipin - the signature phospholipid of the inner membrane that anchors the electron transport chain complexes.

What is SS-31?

By stabilizing cardiolipin, SS-31 improves cristae architecture and electron transport efficiency while reducing electron leak and reactive oxygen species generation at the source. It has been through numerous clinical trials (as elamipretide/Bendavia) for mitochondrial myopathies, heart failure, and Barth syndrome, giving it one of the deepest clinical datasets of any research peptide. Preclinical research covers ischemia-reperfusion injury, neurodegeneration, muscle aging and kidney disease models.

Mechanism of Action (Research Context)

Selectively partitions into the inner mitochondrial membrane and binds cardiolipin, stabilizing cristae curvature and respiratory supercomplex organization. Reduces electron leak and mitochondrial ROS production, improves ATP synthesis efficiency, and inhibits cardiolipin peroxidation-driven cytochrome c release.
