IGF-DES is a growth factor research compound with a molecular weight of 7371.4 g/mol and a research half-life of ~20-30 minutes. IGF-DES (Des(1-3)IGF-1) is a truncated analog of human IGF-1 missing the first three N-terminal amino acids. This small structural change has a large pharmacological effect: IGF-DES binds IGF-binding proteins (IGFBPs) roughly 100-fold less avidly than native IGF-1, leaving far more free, receptor-available peptide in tissue.

What is IGF-DES?

The result is a short-acting but highly potent research tool - reported as several-fold more potent than IGF-1 at the IGF-1 receptor in-vitro, with a plasma half-life of only 20-30 minutes. Research interest centers on localized tissue growth models, muscle repair, and neuronal studies. It sits between native IGF-1 and the long-acting IGF-1 LR3 in the potency/duration trade-off, and its short half-life makes timing and localization central to study design.

Mechanism of Action (Research Context)

Agonist at the IGF-1 receptor (IGF-1R), activating PI3K/Akt and Ras/MAPK pathways driving cell proliferation, protein synthesis and glucose uptake. N-terminal truncation reduces IGFBP binding ~100x, increasing free bioactive fraction; potency at the receptor itself is enhanced relative to native IGF-1.

IGF-DES is a 3-amino-acid N-terminal deletion with a 20-30 minute half-life and very low binding-protein affinity; IGF-1 LR3 has 13 extra N-terminal amino acids plus an Arg3 substitution giving a 20-30 HOUR half-life. DES is short/localized, LR3 is long/systemic.
Removing the N-terminal tripeptide reduces IGF binding protein sequestration by ~100-fold, so far more peptide stays free to activate IGF-1 receptors. In-vitro assays show several-fold higher potency per unit mass.
