Humanin is a mitochondrial research compound with a molecular weight of 2687.2 g/mol and a research half-life of hours. Humanin is a 24-amino-acid peptide encoded within the mitochondrial 16S rRNA gene (MT-RNR2) - one of the first discovered 'mitochondrial-derived peptides', a class of small signaling molecules encoded by short open reading frames in mitochondrial DNA. It was identified in 2001 from a screen for factors protecting neurons against Alzheimer's-related insults.

What is Humanin?
Research has since shown humanin acts as a broad cytoprotective and metabolic signaling peptide: circulating levels decline with age, and higher levels correlate with longevity in several species including humans. Studied areas include neuroprotection, insulin sensitivity and glucose metabolism, cardiovascular protection, and apoptosis regulation. Potent analogs such as HNG (S14G-humanin) are used in research where greater potency is needed. Humanin signals through both intracellular (BAX-binding) and cell-surface receptor (CNTFR/WSX-1/gp130 and FPRL-1) pathways.
Mechanism of Action (Research Context)
Dual signaling: extracellular humanin activates a tripartite receptor complex (CNTFRalpha/WSX-1/gp130) triggering JAK2/STAT3 and AKT survival pathways, and binds FPRL-1/FPR2. Intracellular humanin binds pro-apoptotic BAX and IGFBP3, suppressing mitochondria-mediated apoptosis.
